The genetic variation between two Verticillium dahliae type strains was investigated. The two investigated type strains were different in their colony characteristics; V. dahliae CBS 179.66 possesses white colonies reflecting a defect in microsclerotia formation while V. dahliae var. longisporum CBS 649.85 has dark colonies ensuring the production of microsclerotia. RAPD analyses, using four primers, revealed their great intraspecific variations where a large number of polymorphic bands were generated. RFLP-ITS revealed their interspecific relationship. This study is a basis for a number of molecular research concerning microsclerotia which are the highly resistant and viable structures responsible for the perennation of V. dahliae spp. in soil.
