This study was carried out in Plant Tissue Culture Laboratory at EI-Zohria Botanical Garden, Horticulture Research Institute, Agriculture Research Center during 2007-2008 seasons. The object of this study wa:; to establish a protocol for micropropagation and RAPD analysis for comparing between mother plant and in vitro plant of Pyracantha fortuneana Roem. The data revealed that for sterilization explant, using 40% c1orox for 25 min gave the best result (100% survival) and free contamination explant. Bud break and shoot development were best on full MS establishment medium with (2.0 mgll IBA) which gave the longest shootlet. While the greatest number of leaves was recorded on full MS-medium with (4.0 mglIBA). For shooting behaviour. the medium supplemented with (3.0 mgll BA) for four subcultures gave the greatest number of shootlet. But adding (4.0 or 5.0 mgtl kin) for four subcultures produced the tallest shoodet length. For the number of leaves. culture the explant on MS-medium have (4.0 mgt/ BA) for four subcultures gave the highest number of leaves. Regarding to the rooting behaviour, culturing the shootlet on MS-medium supplemented with (4.0 mg/lBA) produced the highest percentage of rooting, tallest root length and highest number of root. RAPD-based DNA fingerprints gave no evidence of somaclonal variation during the in vitro propagation protocol used. The RAPD profiles of tissue culture-derived plantlets revealed high similarity to mother tree. For acclimatization. the survival percentage was 70%.
