A total number of 216 apparently healthy live Oreochromis niloticus (0. nifoticus) with an average body weight 35 ±5g were used to determine the pathogencity and the virulence of the pathogenic isolates of flavobacterium columnare (F. columnare). Also,the effect of temperature and salinity on the virulence of the pathogenic isolates. The obtained results revealed that, experimentally infected O. niloticus with F. columnare showed loss of escape reflex, swimming near the water surface, erratically movement in water, excessive mucus secretion on body and gills, loss of scales, fin rot especially at caudal and dorsal fins, the site of infection was slightly oedematous and hemorrhagic, severe ulcerative lesions on the body, gills in more advanced cases become severely hemorrhagic and gill rot appear with ulcerative lesions on gill cover, excessive mucous in the gill filament and Kidneys were congested.
