An Egyptian isolate of ToMV was subjected to molecular characterization. The virus was purified from tobacco leaves using differential centrifugation. The electron microscopy of purified vims ptepaIlltion showed the presence of rod-shaped vims-like particles with a model size close to 300 X 17 om. The SOS-polyacrylamide gel electrophoresis (SOS-PAGE) showed a single coat protein band with molecular weight of ahout 17 KDa ToMV coat protein gene (cp) was is0lated and amplified using immunocaptwe reverse transeriptase polymerase chain reaction (IC-RT-PCR) and its size length on agarose gel was found to be 479 bp.
