Summary: The present study aimed to investigate the relationship between sperm chromatin structure, integrity of sperm membrane as well as fertilizing capacity of sperm and quality of frozen buffalo semen. Eight sexually mature buffalo bulls aged 5-6 years were used in the present work. All these animals were clinically normal and free from venereal diseases. The semen samples were collected by A.V. Immediately after collection semen samples that had more than 60% progressively motile sperm were processed (I part semen + 4 parts extender) at 37 'c with a single stepwise addition of citrate and Trisbased extenders) and loading in 0.25 ml PVC ministraws (IVM, L'Aigle - France).
