Subject(s): Online resources: In: Alexandria Journal of Agricultural Research 2007.v.52(3)Summary: In the present work seven isolates of bacteria representing different localities of Menofia Govrnerate -Egypt were collected namely:- Shabin Alkom, TocK. Tala, Monoof El-shohada. Ashmoon, and Quisna. The isolates were obtained from localities characterized with dense rice strow and limited vegetation. This identification was based on the ability ofbacteria to degrade cellulose by Congo &dstain. The enzyme activity was determined by hydrolyzing cellulose. The bacterial isolates were inoculated into LB medium that contained carboxymetlylcellulose (CMC) as the sole corbon source with 3,5 dinitrosalicylic acid (DNS) reagent. then selected the most cellulytic bacterial isolate. The presence ofa clear halo indicates endogiucanase enzyme activity, the presence ofa clear halo arround isolate Nb₄ indicales enzyme activity degradation of CMC more than other isolates, isolate come from a locality characterized by dense rice straw (Monoof). The isolate Nb₄ was identified as Bocillus subtilis by Agriculture &search Center-Egypt, it was Gram positive, aerobic and rodshaped. The maximum extracellular cellulase activity was (60 unit/ml) for the isolale Nb4 while the minimum enzyme activity was (60 unit/ml) for the isolate Nb₇.
