This study was carried out during the period from 2009 to 2012 in Plant Tissue Culture Laboratory of the Hort. Dept., Fac. Agric., Zagazig University to create a protocol for propagating Nemaguard peach rootstock throughout the various stages of tissue culture technique. The obtained results could be summarized as follows: The establishment stage with nodal explants of 1cm length and 2-3 mm. thickness obtained from growing Nemaguard seedlings of two years old proved the efficiency of 2 ppm BA in MS medium for shoots proliferation. During multiplication stage for the shoots of 1cm length obtained from the establishment stage, different concentrations; i.e., 1, 2, 4 or 6ppm ofBA or Kin., in MS medium indicated the efficiency of 4 ppm BA in producing higher number of both shoots (5- 8.5 shoots/ explant) and leaves / shoot. Using NAA and BA at 0,1,2 and 4 ppm each alone or in combinations in MS medium declared that NAA supressed multiplication and produced roots when either NAA was used alone or in combination with BA, whereas 4ppm BA alone still the best growth regulator for multiplication stage. Using adenine sulphate at 10, 20 or 40 ppm alone or combined with 1, 2 or 4 ppm BA did not show any beneficial effect in enhancing more multiplication .
